This covalent binding deactivates the enzyme, halting hydrogen ion transport from parietal cells into the gastric cavity and interrupting the final stage of gastric acid production, which substantially lowers gastric acid concentration. Accordingly, omeprazole delivers powerful, long‑term suppression of gastric acid secretion provoked by diverse stimuli. It suppresses both basal acid output and acid release stimulated by histamine, pentagastrin and vagal excitation. Notably, it also inhibits acid secretion driven by dibutyryl cyclic adenosine monophosphate, a pathway unresponsive to H₂‑receptor antagonists. Such robust and prolonged pharmacological effect stems from its irreversible inhibition against the proton pump; gastric acid secretion resumes only after the human body synthesizes new proton‑pump proteins.
